چکیده
Biochemical markers for Alzheimer disease would be of great value, especially to help in early diagnosis of the disease. The current study has focused on three candidates that have been suggested to be indulged in the pathogenesis of Alzheimer’ disease (AD): Tissue transglutaminase (tTG) activity, β-amyloid42 (Aβ 42) and total T-tau (T-tau). The study included 15 patients with probable AD, 15 patients with probable vascular dementia (VaD) in addition to 10 control subjects without neuropsychiatric diseases. The results of this study revealed that the CSF levels of total T-tau and activity of tTG, were significantly higher in patients with AD when compared with VaD and control. Also, CSF Aβ42 of patients with Alzheimer’s disease were significantly low when compared with vascular dementia, and controls. The sensitivity and specificity of CSF T-tau and Aβ42 were found to be high for differentiation of AD patients from controls, but their specificity against (VaD) is moderate. So, measurement of CSF tTG, T-tau and Aβ42 level in AD patients may provide useful and relatively simple biochemical tests for differentiating Alzheimer’s Disease from vascular dementia .In addition, the diagnostic usefulness of the CSF content of both T-tau and Aβ42 is superior to either measure alone with high sensitivity and specificity for prediction of AD, and can be recommended as an aid to evaluate individuals suspected of having dementia. Biochemical Diagnosis of Alzheimer Disease by Measuring the Cerebrospinal Fluid Ratio of Phosphorylated tau Protein to -Amyloid Peptide42
Background: The antemortem diagnosis of Alzheimer disease (AD) requires time-consuming and costly procedures. Therefore, biochemical tests that can direct the physician rapidly to the correct diagnosis are highly desirable. Measurement of single biochemical markers in cerebrospinal fluid (CSF), such as total tau protein and -amyloid peptide42 (A 42), shows robust alterations that highly correla...

BACKGROUND The aim of this study was to explore the relationship between cerebrospinal fluid Alzheimer's disease (AD) markers and depression in elderly people. METHOD We included subjects with AD as well as persons with subjective cognitive impairment and normal cognition. Depression was assessed with the Cornell Scale for Depression in Dementia, and a cut-off score of >6 was used to define d...
متن کاملComparison of cerebrospinal fluid levels of tau and Aβ 1-42 in Alzheimer disease and frontotemporal degeneration using 2 analytical platforms.
OBJECTIVE To use values of cerebrospinal fluid tau and β-amyloid obtained from 2 different analytical immunoassays to differentiate Alzheimer disease (AD) from frontotemporal lobar degeneration (FTLD). DESIGN Cerebrospinal fluid values of total tau (T-tau) and β-amyloid 1-42 (Aβ 1-42) obtained using the Innotest enzyme-linked immunosorbent assay were transformed using a linear regression mode...
متن کاملEffect of immunotherapy with bapineuzumab on cerebrospinal fluid biomarker levels in patients with mild to moderate Alzheimer disease.
BACKGROUND Given the slow and variable clinical course of Alzheimer disease, very large and extended clinical trials are needed to identify a beneficial clinical effect of disease-modifying treatments. Therefore, biomarkers are essential to prove that an anti-β-amyloid (Aβ) drug candidate affects both Aβ metabolism and plaque load as well as downstream pathogenic mechanisms. OBJECTIVE To eval...
متن کاملThe role of clusterin in amyloid-β-associated neurodegeneration.
IMPORTANCE Converging evidence indicates that clusterin, a chaperone glycoprotein, influences Alzheimer disease neurodegeneration. However, the precise role of clusterin in Alzheimer disease pathogenesis is still not well understood. OBJECTIVE To elucidate the relationship between clusterin, amyloid-β (Aβ), phosphorylated tau (p-tau), and the rate of brain atrophy over time among nondemented ...
